No, and most people don't. The guidebook gives you an order to add them in. Each one does its own job, so you can start with three and build.

Most of it works the first night. Darkness, quiet and nose breathing need no build-up. Magnesium takes a few nights to settle in. If nothing has shifted after 30, that's what the refund is for.
